Accelerating early-stage process development: LC-MS/MS for HCP profiling in a moss-based expression system

Given the lack of commercial HCP assays suitable for its innovative expression system, eleva GmbH – a clinical-stage biopharmaceutical company – initially developed a custom HCP-ELISA with high antibody coverage tailored to its original process. However, substantial process modifications introduced for a subsequent protein candidate led to a significant reduction in assay coverage, necessitating the time-intensive development of a second-generation ELISA.
To maintain project timelines, Eleva simultaneously adopted an LC-MS/MS (SWATH-MS) workflow to identify and quantify individual HCPs. This option enabled a timely, data-driven optimization of the downstream purification process.
